Time progresses on the server as it does in real life. Days begin at 12:00am EST (-5 GMT) and end at 11:59pm EST. However, as a legacy of when the sun still existed, there are technically roughly two day/night cycles per RL day.
For purposes of time-keeping in this cycle, 1 hour equals 27 real-life minutes. However, for the purposes of anything mechanical (spell duration, time until rest, etc.) hours are 5 RL minutes long.
The common calendar used in the setting will be that of Harptos. The following link explains a bit about this particular calendar and includes a small tool players might find useful in determining what the exact date is in game:

Starting Date
The first day of play on the server was November 3rd, 2005 and will be considered Shieldmeet of 1372 in game.
Sanctuary's Calendar
In addition to the standard Forgotten Realms Calendar, it is common in Sanctuary to mark the passage of years since Sanctuary's founding. For example, it is currently Year 161.
Time Skips
Note that there was a several year "time-skip" in between Chapter 2 (EFU:A) and Chapter 3 (EFU:M) and a brief one between Chapter 3 and Chapter 4 (EFU:R).
